What are the primary differences between the iCell Hepatocytes 2.0 and the previous 1.0 version?
The primary differences between iCell® Hepatocytes 2.0 and iCell Hepatocytes 1.0 include:
- Extracellular Matrix Requirements: iCell Hepatocytes 2.0 do not need the Matrigel Matrix formerly needed to support culture of version 1.0, and instead are compatible with a collagen extracellular matrix.
- Healthier Cultures: While our earlier 1.0 version used a development process that generated very good thawed and plated hepatocytes, we identified a slightly different process point for cryopreservation for version 2.0 that generates a healthier culture after thawing despite a longer time to recovery and monolayer development. Flat cobblestone morphology appears by day 4 - 6 post-thaw, and the cells are ready to assay at day 7 post-thaw.
- Additional Data: We have data for iCell® Hepatocytes 2.0 for marker expression via ICC, modulation of the insulin signaling pathways, susceptibility to viral infection (HCB/HBV/malaria), lipid accumulation, NASH modeling, and more. Some of this data (i.e., viral infection) was not possible for the 1.0 version of these cells.
