What protocol do you recommend for plating iCell Cardiomyocytes2 directly onto Axion MEA plates?

Our iCell® Cardiomyocytes2 can be cultured to form a stable electrically and mechanically active syncytium and analyzed as early as 4 days post-thaw using multielectrode array (MEA) technology. MEA technology enables electrophysiological examination and in vitro screening of compound effects by noninvasive, label-free measurements of local field potentials in electrically active cells. Our Application Protocol describes how to handle iCell Cardiomyocytes2 for use on the Maestro MEA system (Axion BioSystems) and provides basic instructions for compound treatments, data acquisition, and analysis.

General protocol recommendations:

  • Assay timing: Day 4 - 8 post-plating following a direct plate dotting method
  • Assay density: 50,000 viable cells in a 5 μl dot (10,000 cells/μl)
  • Assay media: iCell Cardiomyocytes Plating Medium (dot) / iCell Cardiomyocytes Maintenance Medium. Note that cell dots (in Plating Medium) will be gently flooded with Maintenance Medium 1 hour post-dotting with 300 μl per well.
  • Assay ECM: fibronectin (5 μl/well droplet of 50 μg/ml stock solution)
