While our iCell® Cardiomyocytes Plating and Maintenance media for iCell Cardiomyocytes2 contain serum and will expose cells to it during the culture conditions recommended in our User Guide, we have performed a complete medium exchange to serum-free conditions before the assay for incubation lasting overnight (exchange the night before assay) or 4 hours (exchange on the morning of assay) for Maestro (48-well plate, Axion BioSystems) and xCELLigence Cardio RTCA 96-well plate (ACEA Biosystems) applications. We cannot provide support for serum-free incubation beyond 24 hours.

For this serum-free assay medium, we recommend a DMEM-based medium (without glucose) containing 1 mM sodium pyruvate (diluted from a 100x solution) and 10 mM galactose (weighed from powder). Be sure to remove all serum-containing medium from the wells during the exchange, allow at least 4 hours for recovery after medium exchange, and prepare compound dilutions in the serum-free medium.
